Like two molecular locks guarding a treasure, the RCA–CRISPR cascade integrated with SERS establishes sequential checkpoints to unlock ultrasensitive and single-nucleotide-resolved detection of exosomal miRNAs. This layered strategy merges amplification, precision recognition, and optical readout, paving the way for reliable, next-generation cancer diagnostics.
